Antitumor Effect of Recombinant Interferon ¥á-2a in Nude Mice







Abstract
We studied the antitumor effect of recombinant human interferon alph¥á-2a (IFN ¥á-2a) against human chronic myeloid leukemia cell line, KU-812F, which was transplanted in nude mice.
In nude mice the tumor inducing rate of KU-812E cell line (100%) which was selected as the cell line in this experiment was higher than that of K-562 cell line (20%). IFN ¥á-2a treated group showed antitumor effect to max. 1¡¿10^7IU without any toxicity. After the tumor weight reached 100-300§§, the agents were administrated. In the group treated with IFN ¥á-2a 1¡¿10^6IU, disappearance of the tumor was observed and T/C (%) value was reduced to 35%. In the group treated with hydroxyurea 0.5§·/g B.W., marked weight loss was observed. V_r/V_c value of the group was reduced but increased afterward. T/C(%) value of the group administrated with IFN ¥á-2a 5¡¿10^5IU and hydroxyurea 0.25§·/g B.W. was reduced to 76% which was lower than that of the group treated with IFN ¥á-2a 1¡¿10^5IU. The tumor inducing rate was markedly reduced when the mice were inoculated IFN ¥á-2a shortly after the tumor transplantation. In groups administrated with IFN ¥á-2a 5¡¿10^6IU and 1¡¿10^7IU, 40% rejection rate in tumor transplantation was observed.
As a result, We confirmed that recombinant human IFN ¥á-2a can inhibit the proliferation of chronic myelogenous leukemia cell(KU-812F) transplanted in nude mice.
